摘 要:目的:通过对栀子环烯醚萜类进入脑脊液的移行成分分析,阐明栀子治疗中枢神经系统疾病潜在的活性成分。方法:给予犬静脉注射栀子环烯醚萜类成分,并采集3个不同时间点的脑脊液,通过液相色谱-质谱联用(LC-MS)技术检测并鉴定脑脊液中的栀子环烯醚萜类成分,通过MS和核磁共振(NMR)检测余下的栀子环烯醚萜成分。结果:在脑脊液中存在3种栀子环烯醚萜类成分。利用LC-MS分析确认其中1种成分为栀子苷;而采用MS和NMR数据确证的其他2种栀子环烯醚萜类成分分别为去乙酰基车叶草苷酸甲酯和京尼平-β-D-龙胆双糖苷。结论:栀子环烯醚萜类成分中栀子苷、去乙酰基车叶草苷酸甲酯和京尼平-β-D-龙胆双糖苷能够通过血脑屏障进入脑脊液,其透过的机制和药理作用还有待进一步研究。OBJECTIVE: To elucidate the potential active components of Gardeniae jasminoides in the treatment of central nervous system disease by analyzing constituents absorbed into cerebrospinal fluid in iridoid compounds extracted from G. jasminoides METHODS: The iridoid compounds extracted from G. jasminoides were given by intravenous injection in dogs, and cerebrospinal fluid was obtained at three different time points. The G. jasminoides iridoid compounds in cerebrospinal fluid were detected and identified by LC-MS analysis. Remaining G. jasminoides iridoid compounds were determined by MS and NMR. RESULTS: We found 3 G. jasminoides iridoid compounds in cerebrospinal fluid. One was geniposide by LC-MS analysis, others were deaeetyl- asperulosidic acid methyl ester and genipin-1-β-D-gentiobioside rspectively by NMR and MS spectra analysis. CONCLUSION:Geni- poside, deacetyl-asperulosidic acid methyl ester and genipin-1-β-D-gentiobioside can be absorbed into cerebrospinal fluid through blood-brain barrier. The mechanism and pharmacological effect are worthy of further studying.
