摘 要:目的研究山核桃叶提取物体外对人肝癌细胞株HepG2的抑制作用及其可能机理,为进一步开发利用山核桃奠定基础。方法采用系统溶剂法将山核桃叶70%乙醇浸膏分成五个部位,然后通过MTT法观察其对人肝癌细胞株HepG2的抑制作用,并通过吖啶橙染色法和流式细胞术观察其诱导凋亡作用。结果山核桃叶氯仿层和石油醚层对HepG2细胞均具有较强的增殖抑制作用,氯仿层相对较强,IC50为(49.70±4.42)mg.L-1,并可剂量依赖性诱导HepG2发生细胞凋亡,其中100mg.L-1处理组凋亡率达28.0%。结论山核桃叶氯仿层中和石油醚层含有抗肿瘤活性成分,其中诱导细胞凋亡是氯仿层提取物发挥抗肿瘤活性的作用机理之一,值得进一步深入研究。OBJECTIVE To investigate the proliferate inhibition effects and uncover the underlying mechanism of Carya Cathayensis Sarg.leaf extracts(CCSLE) against HepG2 cell lines in vitro,which would pave the way for its further exploitation and utilization.METHODS The leaf of Carya Cathayensis Sarg.was extracted by 70% ethanol,and then partitioned into five sub-fractions.MTT method was carried out to observe the inhibition effect of the fractions;Acridine orange staining method and flow cytometry technique were adopted to analyze the potential inducing cell apoptosis effect of the active fraction.RESULTS The sub-fraction of petroleum and chloroform were verified to be active part of CCSLE,and the later was more effective,with IC50=(49.70±4.42)mg·L-1,which can also dose dependently induce cell apoptosis,as 28.0% apoptosis ratio was reached treating by 100mg·L-1 chloroform part.CONCLUSION Petroleum and chloroform,these two sub-fractions of CCSLE contain antitumor active components;Inducing cell apoptosis is one of the mechanism which the chloroform part exert its antitumor activity;which deserve further exploitation.
