摘 要:目的研究鸡体内细胞因子的基础转录水平。方法利用双重荧光定量RT-PCR(dqRT-PCR)技术,对70日龄的5种主要组织相容性复合体(Major histocompatablity complex,MHC)单倍型无特定病原体(special pathogen free,SPF)鸡群G1、G2、G3、G5和G7的肺组织(包括气管)、胸腺、法氏囊、脾脏和外周血淋巴细胞(PBL)中Th1型(IFN-γ、IL-18)和Th2型(IL-4、IL-10)细胞因子mRNA含量进行定量检测。结果 IFN-γ和IL-4在各鸡群的胸腺中转录水平最高,IL-18和IL-10在法氏囊中最高,Th2型细胞因子在脾脏中最低;初级免疫器官(胸腺、法氏囊)中IFN-γ和IL-10转录水平在G3鸡群中最高。结论 70日龄时Th1型和Th2型细胞因子在SPF鸡的初级免疫器官中转录水平较高,且为制衡状态,胸腺中表现为IFN-γ和IL-4相互制衡,法氏囊中为IL-18与IL-10;不同MHC单倍型鸡体内细胞因子转录水平有所差异,但未达到显著水平。Chicken major histocompatibility complex(MHC) is a genetic complex related to immunity.Chicken line G1,G2,G3,G5 and G7 are five haplotypes belonging to B13,B15,B2,B5 and B19 respectively,confirmed by microsatellite loci of LEI0258 and MCW0371 which located in chicken major histocompatibility complex.In order to study the basic transcription level of cytokines,the five chicken lines were chosen to quantify Th1(IFN-γ,IL-18) and Th2(IL-4,IL-10) cytokines in lungs,thymus,bursa of Fabricius,spleens and peripheral blood lymphocytes(PBL) based on duplex quantitative real-time reverse transcription polymerase chain(dqRT-PCR) method.The results shown that IFN-γ and IL-4 were abundant in thymus,IL-18 and IL-10 were highest in bursa of Fabricius,and Th2 cytokines were the lowest in spleens in the seventy days old chickens.Meanwhile,IFN-γ and IL-10 in the primary immune organs(thymus,bursa of Fabricius) were highest in line G3.In conclusion,Th1 and Th2 cytokines are higher in the primary immune organs of seventy days old chickens,and shown to balance to each other: IFN-γ and IL-4 in thymus,IL-18 and IL-10 in bursa of Fabricius;there are some differences of the transcription level of cytokines among different MHC-B haplotype chickens,but not significant.
