摘 要:目的:通过研究生脉成骨胶囊对激素性股骨头缺血性坏死的股骨和髂骨的间充质细胞的碱性磷酸酶(ALP)和骨钙素(BGP)的变化,探讨其对髂、股骨骨髓干细胞成骨能力的影响。方法:将60只SD雄性大鼠随机分为6组:空白对照组、模型组、生脉成骨预防用药组、阿仑磷酸钠治疗组、丹郁骨康治疗组、生脉成骨治疗组,除空白对照组外,其余大鼠腹腔注射醋酸泼尼松龙,建立激素性股骨头坏死动物模型。造模成功后,各组给予相应的药物或生理盐水进行灌胃。取其股骨及髂骨,收集骨髓细胞悬液进行成骨细胞诱导培养,分别用ELISA法测定ALP、BGP的含量。结果:空白对照组、生脉成骨预防用药组、阿仑磷酸钠治疗组、丹郁骨康治疗组、生脉成骨治疗组股骨髂骨的ALP、BGP含量均显著高于模型组(P<0.05),丹郁骨康治疗组的股骨和髂骨,以及生脉成骨治疗组的股骨ALP含量显著高于阿仑磷酸钠治疗组(P<0.05)。结论:生脉成骨胶囊可以促进激素性股骨头坏死大鼠模型的股骨、髂骨间充质细胞碱性磷酸酶和骨钙素合成,在促进成骨细胞分化成熟方面上显著高于阿仑磷酸钠片。初步认为生脉成骨胶囊治疗激素性股骨头缺血性坏死的作用机理与促进成骨细胞的分化成熟程度以及提高成骨细胞骨蛋白的分泌速度有关。Objective:To evaluate the bone formation ability of the mesenchymal stem cells in iliac bone and proximal femoral bone marrow, by the changes of ALP and BGP effected by Shengmaichenggu capsule in mesencbymal slem cells of lilac bone and proximal femoral bone of SANFH. Methods:60 SD male rats, according lo the random number tablc, were divid ed into 6 groups: the control group, group of preventive medication of Shengmaichenggu, model group, Shengmaichenggu group, Danyugukang group and Alendronate group. Models were established by injecting prednisolone inside abdominal cavity for all the rats, except rats in the normal group. All rats were given appropriate drugs or normal saline by gavsges, after SANFH models were established. Mesenchymal stem cells were executed and cultured from the lilac bone and proxi real bone, BGP and ALP in the mesenchymal stem cells were determined by ELISA Kit. Results:The level of BGP and ALP in the femur and iliac bone in the control group, Shengmaichenggu preventive group, Alendronate group, l)anyugukang treatment group and Shengmaichenggu treatment group were remarkably higher than that in lhe model group(P〈 0.05 ) ; the level of AI.P in the fenmr and lilac bone in Danyugukang treatment group was remarkably higher than that in Alendr onate treatment group(P〈0.05). Conclusion: The level of ALP and BGP in mesenchymal stem cells of femur and iliac bone of SANFH rat's model was increased, after treated by Shengmaichenggu; it can promote the process of osteoblast differcn tiation. The treatment mechanism to ischemic necrosis may bc related to the process of osteoblast differentiation and excretion rate of osteoblast protein caused by Shengmaichenggu.
