摘 要:为了深入了解西藏核桃种质资源的遗传背景和遗传多样性,采用FISH-AFLP技术,筛选9对EcoRⅠ+3和MseⅠ+3引物组合,对我国西藏核桃主产区加查和郎县的11个核桃(JuglansregiaL.)优良单株进行基因组DNA水平检测。结果表明:在获得的1103条可统计条带中,1076条呈多态性,多态性达97.67%;9对引物所检测到的不同位点的有效等位基因数(Ne)范围为1.00~1.99,平均为1.48±0.04;基因多样度(H)为0.00~0.49,平均为0.29±0.02;Shannon信息指数(I)为0.00~0.69,平均为0.44±0.02。本研究分析了西藏核桃优良单株的遗传多样性和亲缘关系,建立了它们的指纹图谱。In order to deeply understand the genetic background and the genetic diversity of Tibet walnut germplasm resources,FISH-AFLP was conducted using 9 EcoRⅠ+3/MseⅠ+3 primer combinations to analyse 11 walnut(Juglans regia L.) advanced trees from Tibet at genomic DNA level.Totally 1 103 AFLP fragments were obtained,in which 1 076 were polymorphic and the average percentage of polymorphic bands(ppb) was 97.67%.The value of effective number of alleles(Ne) ranged from 1.00 to 1.99 with an average of 1.48±0.04;Nei's gene diversity(H) ranged from 0.00 to 0.49 with an average of 0.29±0.02 and Shannon's information index(I) ranged from 0.00 to 0.69 with an average of 0.44±0.02.The genetic diversity and sibship among walnut advanced trees in Tibet were investigated and their fingerprintings were established in the research.
