摘  要:目的建立"三热论"复方(地骨皮、黄连、山茱萸、鬼箭羽)治疗糖尿病有效部位的指纹图谱,为相关制剂工艺的质量控制提供可靠参数。方法采用HPLC法测定10批样品的指纹图谱。色谱柱为Agilent Zorbax C18柱(250 mm×4.6mm,5μm);流动相A为0.5%的甲酸水溶液,B为甲醇,梯度洗脱,90%A~80%A保持5 min,80%A保持20 min,80%A~60%A保持55 min,60%A保持5 min;柱温30℃;体积流量1 mL/min;检测波长254 nm;进样量为10μL。结果 10批不同样品指纹图谱中,指标成分马钱素、小檗碱等相似度高,重现性好。结论本方法简单易行,重现性好,可用于"三热论"复方有效部位的质量控制。AIM To develop an HPLC fingerprint method for detection of effective parts in the Compound Prescription of "Three Heat Theory"(Lycii Cortex,Ramulus Euonymi,Coptidis Rhizoma,Corni Fructus).METHODS An octadecyl bonded Agilent Zorbax C18 colum(250 mm×4.6 mm,5 μm) was used for HPLC.Column temperature was maintained at 30 ℃.Gradient elution was conducted using 0.5% phosphoric acid water solution as the mobile phase A and methanol as the mobile phase B.Elution was started from 90% A to 80% A for 5 min,then 80% A for 20 min,from 80% A to 60% A for 55 min,and at last 60% A for 5 min.Flow rate was 1 mL/min.The detection wavelength of a photodiode-array detection(DAD) detector was set at 254 nm.RESULTS The standard components(loganin,berberine) of CPTHT from ten different origins showed high similarities in their HPLC fingerprints.Results had very good reproducibility.CONCLUSION The HPLC fingerprint method is simple,accurate and easy to perform.It can be employed for the quality control of effective parts in CPTHT.
