雪莲提取物XL-12的主要药效学研究

摘　　要：目的观察雪莲提取物XL-12在抗炎镇痛及免疫方面的药理作用,为其用于治疗关节炎提供依据。方法取昆明小鼠50只,随机分为5组,每组10只,即模型组、阳性对照组、雪莲提取物XL-12小、中、大剂量组;取Wistar大鼠60只,随机分为6组,每组10只,分组如小鼠,阳性对照药为野木瓜片。采用二甲苯致小鼠耳肿胀、冰醋酸致小鼠扭体、角叉菜胶致大鼠足跖肿胀、Freund’s完全佐剂致大鼠原发性、继发性关节炎损害和棉球肉芽肿等模型对照观察雪莲提取物XL-12的抗炎、镇痛等作用,采用MTT法测定对小鼠T、B淋巴细胞增殖的影响,观察其对免疫系统的影响。结果雪莲提取物XL-12可明显抑制完全佐剂所致大鼠原发性和继发性炎症,能抑制二甲苯致小鼠耳肿胀,明显减少冰醋酸致小鼠扭体反应次数,能减轻角叉菜胶致大鼠足肿胀度,明显抑制大鼠棉球肉芽肿。雪莲提取物XL-12在浓度0.13～4 mg/mL范围内对T、B淋巴细胞的增殖均有明显抑制作用,在浓度<0.06 mg/mL时抑制作用均明显减弱。结论雪莲提取物XL-12有良好的抗炎镇痛和调节机体免疫功能的作用。AIM To study the pharmacological effects of Saussurea involucrata extract XL-12 on anti-inflammatory analgesic and immunization and provide the basis for the treatment of arthritis.METHODS Fifty Kunming mice were randomly divided into five groups（10 each group）,model group,positive control group,Saussurea involucrata extract XL-12 small,medium,and high-dose group.Sixty Wistar rats were randomly divided into six groups（10 each group）,like mice group.The positive contrast was adopted Wild Pawpaw Tablet.The anti-inflammatory and analgesic effects were studied on models of oxylene-induced swelling of ear in mice,acetic acid-induced twisting reaction in mice,carageenan-induced voix pedis swelling in rats,the primary and secondary arthritis of rats induced by the complete Freund’s adjuvant and the proliferation of granuloma.The proliferation of T and B lymphocytes in mice was tested by MTT assay.The effect on the immune system was observed.RESULTS Saussurea involucrata extract XL-12 could obviously inhibit the primary and secondary inflammation of rats induced by the complete Freund’s adjuvant,inhibit the ear edema induced by oxylene in mice,decrease the twisting times of mice caused by acetic acid,alleviate voix pedis swelling induced by carageenan in rats,and markedly inhibit the proliferation of granuloma in rats.Moreover,Saussurea involucrata extract XL-12 could significantly inhibit the proliferation of T and B lymphocytes at concentration in the range of 0.13-4 mg/mL,and inhibition markedly reduced at concentrations less than 0.06 mg/mL.CONCLUSION Saussurea involucrata extract XL-12 has the anti-inflammatory analgesic and regulating immune effects.
