摘 要:目的:探讨数字化载银珊瑚羟基磷灰石/聚乳酸(CHA/PLA)抗菌人工骨材料制备方法及其理化、结构性能与细胞毒性。方法:将珊瑚羟基磷灰石(CHA)粉末浸泡不同浓度的硝酸银溶液,通过溶液离子交换法,制备出不同含银量的载银羟基磷灰石,再将不同的载银CHA粉末与聚乳酸(PLA)按一定比例混合,并通过选择性激光烧结快速成形制备出具有特殊形状的数字化载银抗菌人工骨支架材料。采用扫描电镜、红外光谱仪、原子吸收光谱仪对数字化载银CHA/PLA抗菌人工骨进行理化、结构及Ag+含量分析,采用MTT法检测材料的细胞毒性。结果:不同含银量的数字化载银CHA/PLA抗菌人工骨与末载银CHA人工骨的理化、结构基本相似,仍保留天然微孔及晶体结构。MTT法测定浸泡10-2、10-3、10-4、10-5mol/LAgNO3的载银CHA/PLA人工骨材料的细胞毒性分别为3、1、0、0级。结论:采用CHA粉末浸泡不同浓度的硝酸银,再与PLA混合,通过快速成型技术可成功地制备出含银量不同的数字化载银CHA/PLA抗菌人工骨材料,且少量载银量对细胞无毒性。Objective To research the preparative method of the antibiosis material of digital coral hydroxyapatite/polylactie acid (CHA/PLA)artifical bone loading Ag+ ,and study its structural properties and cytotoxicity. Methods Put the powder of coral hydroxyapatite (CHA) immersed in different concentration of AgNO3 solution, through solution ion exchang method, prepare coral CHA loading different quantity of Ag+ , then combine the different powder of CHA loading different Ag+ with polylactic acid (PLA) according to fixed proportion, then prepare the antibiosis support material of digital CHA/PLA artifical bone loading Ag+ with special shape through Selective Laser Sintering of Rapid Prototyping Technology.Use Scanning Elecrton Microscope, Infrared Spectrometer, Atomic Absorption Spectrometer to analyze the chemical and structural properties and Ag+ content of digital CHA/PLA artifical bone loading Ag+, and MTT method was used to evaluate the cytotoxicity. Results The chemical and structural properties of digital CHA/PLA artifical bone loading different quantity of Ag+ were similar with CHA artifical bone of unloading Ag+ ,still remain natural micropore and crystal structure. The cytotoxicity grades of MTr method were 3,1,0 and 0 respectively in according to the artifical bone of 10-2, 10-3, 10-4 and 10-5 mol/L AgNO3. Conclusion Use the powder of CHA immersed in different concentration of AgNO3 solution, then combine with PLA, can prepare successfully the antibiosis material of digital CHA/PLA artifieal bone of loading different quantity of Ag+ by Rapid Prototyping Technology, and little Ag+ has no cytotoxicity.
