摘 要:目的:通过体外筛选方法,优选川芎治疗冠心病的有效成分的提取纯化工艺。方法:培养原代乳鼠心肌细胞,利用连二亚硫酸钠(Na2S2O4)缺氧1 h复氧1 h,损伤心肌细胞制备心肌细胞缺氧-复氧损伤模型来模拟体内缺血再灌注损伤,通过体外筛选方法,优选出川芎治疗冠心病的有效成分的最佳提取和纯化工艺。结果:优选出川芎的最佳提取工艺为使用6倍量的水,提取2次,每次1 h;其纯化工艺为使用15 ml HPD-300树脂,取9 BV的0.5 g/ml的川芎提取液上样,以5 BV的50%醇洗脱。结论:通过体外筛选的提取纯化工艺可信度高,为开发研究心脑血管疾病的药物提供了依据。Objective: To select the best extraction and purification process of active ingredients of Chuanxiong which could treat the coronary heart disease through screening methods in vitro.Methods: Primary cultured neonatal rat cardiomyocytes and sodium hydrosulfite(Na2S2O4) were used for hypoxia 1 h and reoxygenation 1 h to form the cardiomyocyte hypoxia-reoxygenation injury model to simulated in vivo ischemia-reperfusion injury.Screening methods in vitro were used to select the best extraction and purification process of active ingredients of Chuanxiong which could treat coronary heart disease.Results: The best extraction process selected was that,Chuanxiong was extracted for 2 times,1 h for each time with 6 times the amount of water;while the purification process was that,15 ml HPD-300 resin was used,9 BV of 0.5 g/ml of the Chuanxiong extract and 5 BV of 50% ethanol was applied to elute.Conclusions: The technology of extraction and purification through screening methods in vitro has high reliability,and provides a basis for the development of new drug that treat cardiovascular disease.
