从猪毛囊中快速提取基因组DNA的有效方法

摘　　要：采用酚仿抽提法提取猪毛囊中的基因组DNA,并以此为模板进行猪骨形态发生蛋白7基因(BMP7)片段的扩增鉴定,优化建立一种快速、简单提取动物基因组DNA的方法。琼脂糖凝胶电泳检测结果表明,从猪毛囊中提取的基因组DNA纯度、质量浓度较高,能满足PCR扩增需要。提取DNA所用猪毛囊数量以10根为好。用毛囊提取动物基因组DNA简便、经济,能够在短期内获得群体基因组DNA,且不会对动物造成损伤。In this paper,the DNA of pig hair-follicle was extracted by the phenol and chloroform extraction method,and using it as a template for the amplification identification of BMP7 gene fragment,in order to optimize and establish a fast and easy way of DNA extraction from pig hair-follicle,The agarose gel electrophoresis detection showed the purity and concentration of DNA obtained from hair-follicle was higher,and it could satisfy the need of PCR.The ideal number of pig hair-follicle was 10.It is a convenient and inexpensive way to extract pig hair-follicle which can not cause injury to animals,and can obtain population genomics DNA in a short period.
