摘 要:为获得羊痘病毒P32基因生物信息学资料,扩增山羊痘病毒贵州LD株的P32基因,并进行克隆、测序与序列分析;以P32基因的推导氨基酸序列为材料,应用计算机技术和生物学软件预测P32蛋白的跨膜结构、亲水性、抗原指数、柔韧性、表面可及性和二级结构,综合评价P32蛋白的抗原表位。结果表明,羊痘病毒P32基因核苷酸和氨基酸同源性分别达到97.8%和95.6%以上;P32蛋白为含有一个跨膜结构的膜蛋白;肽段227-251区域的各种预测指数均高,推断该区域为一个潜在的优势抗原表位。数据分析显示,P32蛋白具有较强的抗原性,该结果将为GPV P32基因工程疫苗的研究和体外表达产物的应用提供依据。In order to get the information of capripoxvirus P32 gene,we amplified the P32 gene by PCR method from nucleic acid sample of goat poxvirus LD strain isolated in Guizhou.The PCR product was inserted into the pMD18-T vector and the gene sequence of the recombinant plasmid has been determined.The nucleotide sequence and deduced amino-acid sequence of goat poxvirus P32 gene were respectively compared with the relative sequence published in GenBank to predict the transmembrane structure,hydrophilicity,antigenic index,flexible regions,surface probability plots and secondary structure of P32 protein.The results indicated that the homology of nucleotide and amino acids exceeded 97.8% and 95.6% respectively;The P32 protein was a kind of membrane protein which had a transmembrane structure;The 227-251 segment was perhaps a larvaceous dominant epitope where all kinds of prediction index were higher.The research analytic data displayed the P32 protein had high antigenicity which will provide theoretical evidence for the research of genetically engineering vaccine and the application of P32 protein.
