摘 要:目的观察黄芪甲苷对兔脂肪来源的间充质干细胞(ASCs)体外增殖的作用。方法提取实验家兔ASCs,培养之后,设实验组和对照组,实验组加入不同浓度黄芪甲苷溶液,对照组加常规培养液,用流式细胞仪观察细胞周期,用四甲基偶氮唑盐(MTT)法测定细胞的增殖率,比较细胞周期和细胞增殖率。结果实验组和对照组培养24 h后,实验组增殖期细胞为72.9%,明显高于的对照组的61.2%(P<0.01);48 h后观察,黄芪甲苷浓度在0.019 5 mg/L时,对ASCs无明显的促进增殖作用(P>0.05);而黄芪甲苷浓度在0.039 1~10 mg/L范围时,对ASCs均有明显的促进增殖的作用(P<0.01)。结论黄芪甲苷对ASCs体外增殖有明显的促进作用。Purpose To investigate the effects of astragaloside on cell cycle and proliferation in vitro of rabbit adipose-derived mesenchymal stem cells(ASCs). Methods ASCs were obtained from an experi- mental rabbit and cultured. An experimental group and a control group were designed. Various concen- trations of astragaloside were added to the culture fluid in the experimental group, while a conventional culture method was employed in the control group. The distributions of the cell cycles were measured by FCM and the proliferation rates of ASCs by MTT. Results After 24 hours of culture, the number of the cells during the proliferative phase for the experimental group was 72.9% , higher than that for the control group by 61.2% (P 〈 0.01 ). After 48 hours, it is suggested that astragaloside at the concentration of 0. 019 5 mg/L did not promote proliferation obviously(P 〉0.05) , but at 0. 039 1-10 mg/L, there were promotion effects on proliferation of ASCs obviously( P 〈 0.01 ). Conclusion Astragaloside has significant promotion effects on the proliferation of rabbit ASCs in vitro.
