摘  要:为分析荷斯坦公牛朊蛋白基因(PRNP)多态性与精子活力的关系,并对其抗病性进行评估,选育抗病公牛,本实验以公牛精液为样品,研究PRNP基因中12 bp、23 bp和24 bp 3个片段的插入/缺失多样性、基因mRNA转录水平及其与精子活力等指标的关系。结果表明,12 bp和23 bp在群体中均得到3种基因型,24 bp只有2种基因型。不同基因型群体的mRNA转录水平存在显著差异,而基因型与精液产量和活力等指标存在相关性,抗病力和精子活力指标存在负相关。本研究中3个片段的插入-缺失多样性可以作为公牛选育的辅助标记。In this study, the polymorphisms of bovine prion protein gene (PRNP) and their relationship on production performances of sperm in Holstein bull were analyzed to select the disease resistant bull. The PRNP mRNA levels and its polymorphisms of 23 bp in-del, 12 bp in-del and 24 bp in-del were detected and their relationships on sperm motility were analyzed. Results showed that polymorphisms at the 23 bp and 12 bp exhibited 3 genotypes including in-in, in-del and del-del. While the polymorphisms at 24 bp fragment displayed 2 genotypes such as in-in and in-del. The PRNP mRNA transcript levels had significant difference between different genotype flocks, and the different levels had effects on sperm motility and ejaculation volume. However, it was also found that there was a negative relationship between the resistance and production performance in Holstein bull. The results in this study could be used as molecular markers for bull selection.
