摘  要:目的观察白藜芦醇对体外培养的人外周血单个核细胞(PBMCs)分泌白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6),单核细胞趋化蛋白-1(MCP-1)及对细胞因子信号抑制因子1和3(SOCS1/3)表达的影响。方法实验分5组,分别用0、3.13、6.25、12.5、25μg/mL的白藜芦醇作用于PBMCs,用实时荧光定量PCR、ELISA实验测定不同浓度白藜芦醇对PBMCs分泌IL-1β、IL-6及MCP-1的mRNA和蛋白的表达,测定SOCS1/3mRNA的表达。结果与对照组相比,应用白藜芦醇后PBMCs分泌IL-1β、IL-6及MCP-1的量减少,SOCS1/3的量增多。在蛋白水平,白藜芦醇浓度为25μg/mL时,IL-1β及IL-6表达明显减少(P<0.05),尤其浓度为6.25μg/mL到25μg/mL时,MCP-1的表达明显减少(P<0.05)。在mRNA水平,各组的IL-1β、IL-6及MCP-1表达均明显降低(P<0.05),而SOCS1/3表达均明显增高(P<0.05)。结论白藜芦醇在剂量依赖性抑制PBMCs促炎性细胞因子IL-1β、IL-6及MCP-1产生的同时,还可以促进免疫负调控分子SOCS1/3的mRNA表达,这可能是白藜芦醇的抗炎作用及作用机制之一。Objective To explore the effects of resveratrol on expressions of interleukin-1(IL-1β),interleukin-6(IL-6),monocyte chemotactic protein-1(MCP-1) and suppressor of cytokine signaling1/3(SOCS1/3) in cultured human peripheral blood mononuclear cells(PBMCs).Methods Cultured PBMCs were treated with different concentrations of resveratrol(0,3.1,6.25,12.5,25μg/mL).Expression of IL-1β,IL-6 and MCP-1 at both mRNA and protein levels,SOCS1/3 at mRNA level were determined using real-time quantitative PCR and ELISA.Results Compared with the control group(0μg/mL),expression of IL-1β,IL-6 and MCP-1 were significantly suppressed by resveratrol,while expression of SOCS1/3 were increased(P0.05).The levels of IL-1β and IL-6 proteins expression significantly decreased at the concentration of 25μg/mL(P0.05),and the level of MCP-1 protein expression decreased in groups from 6.25μg/mL to 25μg/mL(P0.05).IL-β,IL-6 and MCP-1 mRNA expression were significantly suppressed while SOCS1/3 mRNA expression was highly induced by resveratrol at all groups(P0.05).Conclusion Resveratrol can inhibit expression of the pro-inflammatory cytokines IL-1β,IL-6,MCP-1 and induce expression of SOCS1/3 in PBMCs at a dose dependent manner,which further indicates its anti-inflammatory function in pre-clinical studies.
