摘 要:目的观察金雀异黄素在不同钙浓度条件下对大鼠肠系膜动脉平滑肌环单磷酸鸟苷(cyclicguanosine monophosphate,cGMP)水平的影响。方法将成年雄性SD大鼠随机分为实验组(金雀异黄素100μmol/L)和对照组,分别给予下述条件干预:钙离子浓度为0、0.6、1.2和1.8mmol/L,乙二醇-双(2-氨基乙醚)四乙酸[Ethylene glycol-bis(2-aminoethylether)-N,N,N’,N’-tetraacetic acid,EGTA]2mmol/L,硝苯地平10μmol/L,毒胡萝卜素内酯(thapsigargin)2μmol/L和1,2-双(2-氨基苯氧基)乙烷-N,N,N’,N’-四乙酸四(乙酰氧甲基)酯[1,2-Bis(2-aminophenoxy)ethane-N,N,N’,N’-tetraacetic acid tetrakis(acetoxymethyl ester),BAPTA-AM]20μmol/L。采用酶免疫分析试剂盒测定环单磷酸鸟苷(cGMP)含量。结果实验组cGMP含量在低钙浓度组(0、0.6、1.2mmol/L)较在正常钙浓度组(1.8 mmol/L)降低(P<0.01),并随钙浓度下降而降低;对照组cGMP含量低钙组与正常钙浓度组间无明显差异。EGTA和硝苯地平实验组cGMP含量明显升高(P<0.01),毒胡萝卜素内酯、BAPTA-AM实验组cGMP含量明显降低(P<0.01)。结论在正常细胞外钙至零钙水平的不同钙浓度条件下金雀异黄素对大鼠肠系膜动脉平滑肌cGMP的含量影响不同,随细胞外钙浓度降低cGMP含量减少,金雀异黄素在EGTA、硝苯地平存在下使cGMP含量明显增加,然而,在毒胡萝卜素内酯、BAPTA-AM的存在下使cGMP含量下降。Objective To observe the effects of genistein on the cGMP levels of rat mesenteric artery in different calcium conditions.Methods Adult male SD rats were randomly divided into experimental(genistein 100μmol/L)and control groups,which were incubated with calcium buffers(0,0.6,1.2,1.8mmol/L),EGTA 2mmol/L,nifedipine 10μmol/L,thapsigargin 2μmol/L and BAPTA-AM 20μmol/L respectively.cGMP levels were detected by Enzyme Immunoassay.Results Compared to the group with normal calcium concentration,cGMP levels in genistein groups with lower calcium concentrations were significantly decreased(P〈0.01).They became lower with calcium concentration decreasing.There were no significant differences of cGMP levels among control groups with different calcium concentrations.EGTA and nifedipine increased cGMP levels in genistein groups significantly(P〈0.01),while thapsigargin and BAPTA-AM decreased those in experimental groups(P〈0.01).Conclusion The effects of genistein on the cGMP levels of rat mesenteric artery in different calcium conditions are different.As calcium concentration decrease,cGMP levels of genistein groups decrease.Genistein resultes in higher levels of cGMP under existence of EGTA and nifedipine,however,lower when thapsigargin and BAPTA-AM exist.
