牛蒡子苷对糖尿病肾病大鼠肾小球滤过屏障损伤的保护作用

摘　　要：目的探讨牛蒡子苷对糖尿病肾病(DN)大鼠肾小球滤过屏障中nephrin、podocin和乙酰肝素酶(HPSE)表达的影响。方法将SD大鼠制备成DN实验模型,随机分为模型组、牛蒡子苷小、大剂量组(ig 40、60 mg.kg-1.d-1)正常对照组。8周后观察各组血糖、24 h尿蛋白定量及肾脏指数,光镜下观察肾组织的病理改变,采用免疫组化和Real time-PCR检测肾皮质中nephrin、podocin、HPSE蛋白和mRNA的表达。结果牛蒡子苷可降低DN大鼠尿蛋白的排泄,降低肾脏指数,改善肾脏病理,使nephrin、podocin蛋白和mRNA的表达增加,HPSE蛋白的表达降低。结论牛蒡子苷对DN大鼠具有一定的肾脏保护作用,其作用机制可能与调控肾小球滤过屏障中相关蛋白的表达有关。OBJECTIVE To investigate the effect of arctiin on the expression of nephrin,podocin and heparanase（HPSE）in the filtration barrier of kidney glomerulus in the diabetic nephropathy rats.METHODS The diabetic nephropathy model was induced by Streptozotocin.The rats were randomly divided into three groups： diabetic nephropathy group,arctiin group of low dose（the dosage of 40 mg·kg-1·d-1 for intragastric administration） and arctiin group of high dose [the dosage of 60 mg·（kg·d）-1 for intragastric administration].At the same time,normal control group was set up.8 weeks later,the level of blood glucose,24 h proteinuria and kidney weight/body weight we remeasured,the renal pathomorphology change with light microscope was examined,and the expression of nephrin,podocin and HPSE in renal tissue with immunohistochemical technique and real time-PCR were detected.RESULTS Arctiin could decrease the proteinuria excretion and the kidney index,ameliorate the renal pathomorphology,enhance the expression of nephrin and podocin,and decrease the expression of HPSE in diabetic nephropathy rats.CONCLUSION Arctiin can protect the protein and mRNA protein kidney injury in diabetic nephropathy rats.The mechanism may be related to the control of the expression of correlated protein molecules in filtration barrier of kidney glomerulus.
