摘 要:目的了解重组生长激素(rhGH)及γ-干扰素(IFN-γ)对肝硬化蛋白合成代谢的影响。方法采用Proctor改良法,建立CCl4致大鼠肝硬化模型,合格模型随机分为空白对照组(NS,1 mL.d-1,sc)、阳性对照组(马洛替酯100 mg.kg-1.d-1,ig)、rhGH组(rhGH 0.8 IU.kg-1.d-1,sc)、rhGH+IFN-γ组(rhGH 0.8 IU.kg-1.d-1,sc),自第14天开始同时sc IFN-γ2×104IU.d-1,分别于实验第7、14、28天处死实验大鼠。观察大鼠体重的变化、尿氮排除量,检测肝脏白蛋白的mRNA,用凝胶扫描仪扫描测定各电泳带的光密度值。结果 rhGH组体重增加量高于空白对照组及阳性对照组(P<0.05);rhGH组氮排出量低于空白对照组及阳性对照组(P<0.05);rhGH组及rhGH+IFN-γ组Alb mRNA定量均高于空白对照组及阳性对照组(P<0.05),rhGH组及rhGH+IFN-γ组间无统计学差异(P>0.05)。结论外源性rhGH可促进肝硬化大鼠的肝细胞中白蛋白基因表达、mRNA和白蛋白的合成,提高血中白蛋白水平,增加体重,减少尿氮排出,促负氮平衡的恢复。加用IFN-γ后不影响rhGH促进肝硬化大鼠肝脏白蛋白基因的表达。OBJECTIVE To investigate the effect of recombinant human growth hormone(rhGH) and interferon gamma(IFN-γ) on the hepatic protein synthesis and metabolism in the carbon tetrachloride(CCl4)induced cirrhotic rats in vivo.METHODS Cirrhotic models were established by Proctor's method and randomized into four groups,which were treated with normal saline(NS) 1 mL ·d-1 sc,malotilate 100 mg·kg-1·d-1 ig,rhGH 0.8 IU·kg-1·d-1 sc or rhGH 0.8 IU·kg-1·d-1+IFN-γ 2×104 IU·d-1 sc,respectively.Experimental animals were sacrificed at the seventh day,the fourteenth day and the twenty-eighth day.Weight change and urea nitrogen excretion were analysed,hepatic albumin mRNA was detected by recording the optical density using a gel scanner.RESULTS rhGH obviously promoted weight vs the blank control group(P0.05) and malotilate group(P0.05).rhGH decreased urinary nitrogen excretion vs the blank control group(P0.05) and malotilate group(P0.05),rhGH and rhGH+IFN-γ promoted the synthesis of albumin mRNA and plasma albumin,vs the blank control group(P0.05) and malotilate group(P0.05),and rhGH group vs rhGH+IFN-γgroup(P0.05).CONCLUSION Exogenous rhGH may promote albumin gene and mRNA expression as well as weight gain and decrease the urinary nitrogen excretion of CCl4 induced cirrhotic rats,the IFN-γ may not affect rhGH's promotive effect on albumin gene expression.
