摘 要:目的从银杏提取物中分离纯化黄酮苷元,寻找分离提纯银杏黄酮苷元的最优条件。方法对银杏提取物水解液的酸度、水解液的料液比、水解时间、水解液醇度、水解温度、萃取溶剂、萃取条件以及黄酮苷元的结晶进行研究。结果确定了最佳水解液为醇-盐酸为8∶2、最佳固液比为(g.mL-1)1∶50、水解时间2 h、水解温度60℃、萃取溶剂为乙醚-乙酸乙酯(3∶7)混合萃取,回收溶剂后采用甲醇反复结晶即可得90%以上的黄酮苷元晶体。结论采用酸水解、乙醚和乙酸乙酯混合溶剂回流萃取、甲醇重结晶的方法从银杏叶提取物中分离纯化出90%的银杏黄酮苷元的工艺可行。OBJECTIVE To separate and puriy flavonoid aglycones from the extracts of ginkgo biloba leaves,and the optimal conditions for separation and purification were obtained.METHODS The acidity ratio of GBE weight to solvent volume,time of hydrolysis time,alcohol degree,hydrolysis temperature,extraction solvent and conditions,and the crystallization of flavonoid aglycones of the extracts were studied.RESULTS The optimized procedure was as follows:ratio of alcohol to HCl was 8:2,ratio of GBE weight to solven t volume(g·mL-1)was 1:50,hydrolysis for 2 h at 60 ℃,the extraction solvent was the ether and ethyl acetate(3:7)recrgstallization using methanol.90% of the flavonoid aglycones could be obtained by optimized process.CONCLUSION It is feasible to obtain 90% of the ginkgo flavonoid aglycones from the ginkgo biloba leaves using the process mentioned above.
