摘　　要：目的:采用柱前衍生化HPLC法测定铁皮石斛茎和叶多糖中甘露糖的含量。方法:乙醇脱脂,沸水提取得到的多糖,用盐酸水解为单糖,水解产物加入1-苯基-3-甲基-5-吡唑啉酮(PMP)进行衍生化,采用RP-HPLC法测定多糖中甘露糖的含量。结果:茎和叶中多糖主要是由甘露糖和葡萄糖组成,茎中甘露糖含量比叶中高。结论:铁皮石斛茎和叶多糖中甘露糖含量有较大差异,应分别入药。Objective：To determine the contents of mannose in polysaccharide from the stems and leaves of Dendrobium officinale by pre-column derivation-HPLC.Methods：After medicinal Dendrobium was skimmed by ethanol,the polysaccharides were extracted with hot water and hydrolyzed with HCl.Then,the monosaccharides were derivated by 1-Phenyl-3-methyl-5-pyrazolone（PMP）.The contents of mannose in polysaccharide were carried out by HPLC.Results：Polysaccharide in the stems and leaves of Dendrobium candidum.were composed of mannose and glucose.The contents of mannose in stems is higher than that of leaves.Conclusions：The contents of mannose in stems and leaves were different remarkably.They are used as medicines respectively.
