摘 要:目的:采用柱前衍生化HPLC法测定铁皮石斛茎和叶多糖中甘露糖的含量。方法:乙醇脱脂,沸水提取得到的多糖,用盐酸水解为单糖,水解产物加入1-苯基-3-甲基-5-吡唑啉酮(PMP)进行衍生化,采用RP-HPLC法测定多糖中甘露糖的含量。结果:茎和叶中多糖主要是由甘露糖和葡萄糖组成,茎中甘露糖含量比叶中高。结论:铁皮石斛茎和叶多糖中甘露糖含量有较大差异,应分别入药。Objective:To determine the contents of mannose in polysaccharide from the stems and leaves of Dendrobium officinale by pre-column derivation-HPLC.Methods:After medicinal Dendrobium was skimmed by ethanol,the polysaccharides were extracted with hot water and hydrolyzed with HCl.Then,the monosaccharides were derivated by 1-Phenyl-3-methyl-5-pyrazolone(PMP).The contents of mannose in polysaccharide were carried out by HPLC.Results:Polysaccharide in the stems and leaves of Dendrobium candidum.were composed of mannose and glucose.The contents of mannose in stems is higher than that of leaves.Conclusions:The contents of mannose in stems and leaves were different remarkably.They are used as medicines respectively.
