改良CTAB法对西南牡丹总DNA提取工艺的优化  被引量:4

摘  要:为筛选适宜西南牡丹品种基因组DNA的提取方法,通过4因素3水平的正交试验,对提取牡丹基因组DNA的CATB法进行优化。结果表明:提取西南牡丹品种DNA改良的CTAB法的最佳配方为2%的CTAB、2%的PVP、0.5%β-巯基乙醇,三氯甲烷和异戊醇(24∶1)抽提2次,在此条件下,提取的基因组DNA浓度在712.0~780.5ng/μL,A260/A280比值在1.812~1.823,DNA条带清晰,无拖尾现象。说明,利用该改良的CATB方法,可获得高纯度的牡丹DNA。An orthogonal test was conducted to screen an optimum method for genomic DNA extraction from southwest P.suffruticosa cultivars.And the CTAB method of the genomic DNA extraction from P.suffruticosa was optimized as well.The results showed that the best fomula of the optimized CATB method was 2% CTAB,2% PVP,0.5% β-mercaptoethanol,twice extraction by trichloromethane and octanol(24∶1).Under these conditions,the concentrations of DNA were 712.0~780.5ng/μL.The values of A260/A280 were 1.812~1.823,and all the bands of the DNA were both tight and distinct.DNA of P.suffruticosa with high purity could be obtained by using this optimized CATB method.
