摘 要:证实白细胞介素6(IL-6)对幼年大鼠垂体生长激素(GH)、泌乳素(PRL)及卵泡剌激素(FSH)分泌之影响,实验分体外与体内两部分进行.体外实验采取幼鼠垂体组织块培养法,观察人重组IL-6(rhIL-6)对垂体组织每天分泌GH,PRL及FSH的影响及不同浓度rhIL-6的短时效应.体内实验将不同剂量rhIL-6静注幼鼠体内,观察静注后45min,4h及24h血浆GH,PRL和FSG水平的变化.结果:(1)rhIL-6使体外培养的幼鼠垂体组织每天GH的分泌明显高于对照组(P<0001),促GH分泌效应以培养第5d最强,而显著抑制每天PRL的分泌(P<0005),培养第1d作用最强.(2)不同浓度rhIL-6与体外培养的幼鼠垂体组织作用4h,rhIL-6500U与1000U组均使GH分泌明显高于对照组(P<0001),rhIL-61000~6000U各组均显著抑制PRL分泌,呈较明显的剂量-效应关系.体外实验各处理组FSH分泌量与对照组比较无差异(P>005).(3)静注rhIL-605μg及25μg在45min,4h及24h后血浆GH,PRL及FSH的水平与对照组比较均无差异(P>005).实验表明IL-6参与调节垂体前叶激素之分泌。To determine the effects of interleukin ( IL-6) on release of growth hormone(GH), prolactin (PRL) and Follicle stimulating hormone (FSH) by pituitary of immature rats, the study ws divided into two parts. In vitro: To observe the effects of human recombinant IL-6 (rhIL-6) on daily secretion of GH, PRL and FSH by pituitary tissues and the short-effects of various concentration of rhIL-6. In vivo: Treated rats were intravenously administered various doses of rhIL-6, and plasma GH,PRL and FSH were measured at the 45 minutes, 4 and 24 hours after injection. Results: (1) Daily release of GH from pituitary fragments incubation with rhIL-6 ws significantly higher than that of the controls ( P <0 001) , and the peak value of GH was on the fifth day of culture, but daily release PRL of rhIL-6 groups was markedly lower than that of the controls( P <0 005), and the strongest inhibitory effect of rhIL-6 was observed on the first day of culture. (2) The significantly release of GH from Pituitary fragments incubation with 500 units and 1 000 units of rhIL-6 for 4 hours was observed, but higher concentration of rhIL-6 failed to stimulate significantly release of GH; rhIL-6 significantly inhibited the secretion of PRL throughout a concentration range of 500-6 000 units in a dose of dependent manner. In vitro the secretion of FSH of all treated groups was no different in comparison with that of controls. (3) both 0 5 μg and 2 5 μg of rhIL-6 were ineffective to change plasma GH, PRL and FSH in all treated groups at the 45 minutes, 4 and 24 hours after injection. Conclusion: The experiment suggested that IL-6 regulated the secretion of anterior pituitary hormones, and provided the certain theoretical evidence in demonstration of the effects of IL-6 on neuroendocrine immumonetwork.
