大丽轮枝菌分泌蛋白激发子的分离纯化及生物功能研究

摘　　要：利用硫酸铵沉淀、KTA explorer 10蛋白纯化仪、非变性电泳、割胶电洗脱等方法,从大丽轮枝菌(Verticilliumdahliae)发酵液中分离纯化出一种蛋白激发子,经SDS-PAGE电泳检测单一条带,相对分子量为20 kD。该蛋白激发子能够诱导烟草的过敏反应,处理6 h后,处理部位出现水渍状,24 h后出现坏死斑。该激发子可以诱导烟草细胞在较短时间内产生防卫反应信号分子H2O2和NO,并引起活性氧爆发。The protein elicitor from the culture filtrate of cotton Verticillium wilt pathogenic fungus,Verticillium dahliae,was isolated and purified through the procedures of ammonium sulfate precipitation,KTA Explorer 10 Protein Purification System,native-PAGE,corresponding gel cut and electroelution which appeared as a single band in SDS-PAGE gel with molecular mass 20 kD.Infiltrated with this elicitor induced a hypersensitive response in tobacco leaves.Treated post 6 h,the tissue became water damageable,and 24 h after treated,the tissue became necrosis with browning and complete drying.The protein elicitor activated signal moleculars production included hydrogen peroxide（H2O2）and nitric oxide（NO）,and induced reactive oxygen species（ROS）burst in short time.
