重组人肿瘤坏死因子α(rhTNFα)衍生物的纯化及检定

摘　　要：采用热处理、盐析和离子交换层析技术对新型人肿瘤坏死因子α衍生物 (命名为hTNFD)蛋白进行了纯化 ,纯化后的纯度大于 96 % ,回收率为 43.8% ,纯化倍数为 6 .4倍。经L92 9细胞测活结果表明 ,hTNFD的比活可达 1.0 1× 10 1 0 U mg ,比原型TNFα活性提高 46 5倍 ,整个纯化工艺简单、稳定 ,易于放大 ,显示出较好的临床应用前景 ,A novel tumor necrosis factor α(hTNF α)derivative named hTNFD was purified by heating treatment,salting out and ion exchange chromatography.The purity of the derivative after being purified reached above 96%,and the recovery rate and purification fold of it were 43.8% and 6.4, respectively.The detection result of L929 cell activity showed that,compared with hTNFα,the specific activity(1.01×10 10 U/mg) of hTNFD increased by 465 fold.The purification process was simple,stable and easy to be scaled up.
