摘 要:本工作用红藻氨酸(kainicacid,KA)癫痫模型,经蝎毒处理后观察大鼠癫痫发作的行为变化并检测大鼠海马内GABA免疫反应样物质(γaminobutyricacidimmunoreactivity,GABAIR),对国产东亚钳蝎(ButhusmartensiiKarsch,BmK)粗毒抗癫痫反复发作的细胞机制进行初步探讨。KA癫痫大鼠经蝎毒处理3周后,与实验对照组相比,能明显减轻癫痫发作行为。GABA免疫组化的实验显示,用KA3周后,实验对照组大鼠与空白对照组相比,腹侧海马尤其是海马门区GABA免疫反应阳性神经元数目明显减少,免疫染色强度明显降低。实验给药组大鼠8例中,有6例腹侧海马门区GABA免疫反应阳性神经元数目有明显增加,显著高于实验对照组和空白对照组。各组大鼠背侧海马GABA免疫反应阳性神经元数目及免疫染色强度均未见明显变化。上述结果表明,蝎毒能选择性防止癫痫敏感大鼠腹侧海马GABA能中间神经元的损伤,并使GABA的释放量增加。提示此效应很可能是蝎毒抗癫痫反复发作的重要机制之一。In the present study, the effect of scorpion venom (SV) on epileptic susceptibility and its neurochemical mechanisms were investigated. After SV or normal saline (NS) administration for three weeks in epileptic rats, epileptic seizures were much more decreased in kainic acid (KA)+SV group than those of KA+NS group. GABA immun^ocyto^chemistry showed that both (γ aminobutyric acid immuno^re^activity (GABA IR) density and the number of GABA positive neurons were markedly decreased in the ventral hippocampus, especially in hilus of KA+NS group as compared with NS+NS group. However, in the group treated with SV for three weeks, the number of GABA positive neurons and the density of GABA IR were greater increased than those of both KA+NS group and the NS+NS group. The number of GABA positive neurons and the density of GABA IR were unchanged in the dorsal hippocampus of all the four groups. The above results suggest that SV may selectively prevent loss of GABAergic interneurons in the ventral hippocampus of rats susceptible to epileptic seizures and increase the release of GABA as an antiepileptic mechanism.
