摘  要:目的:探讨降糖补肾方改善胰岛素抵抗,治疗糖尿病的作用机制。方法:制备糖尿病大鼠模型,葡萄糖氧化酶法检测空腹血糖;免疫印迹检测InsR磷酸化、IRS-1酪氨酸磷酸化蛋白活性的变化。结果:空腹血糖≥11.1mmol/L为糖尿病大鼠成模标准,成模大鼠随机纳入研究。降糖补肾方能降低糖尿病大鼠的空腹血糖,降糖补肾方高剂量组降糖效果与阿司匹林组相当(P>0.05);免疫印迹法检测结果显示:模型组糖尿病大鼠肌肉组织InsR的磷酸化水平下降,与空白对照组比较有明显差异(P<0.01),降糖补肾低剂量组与阿司匹林组相比P>0.05,说明在改善糖尿病大鼠肌肉组织InsR磷酸化水平方面这两组效果相当。模型组糖尿病大鼠肌肉组织IRS-1的酪氨酸磷酸化下降,与空白对照组比较有明显差异(P<0.01),阿司匹林组、降糖补肾高剂量组与空白对照组相比(P>0.05),说明阿司匹林组、降糖补肾高剂量组能显著改善IRS-1酪氨酸磷酸化水平,两组效果相当(P>0.05)。结论:降糖补肾方一定程度上能增加糖尿病大鼠肌肉组织InsR磷酸化、IRS-1酪氨酸磷酸化蛋白表达,这可能是降糖补肾方抑制炎症信号通路的传导而发挥其改善胰岛素抵抗,治疗糖尿病的机制之一。Objective:To investigate the mechanism of Jiangtang Bushen Prescription in improving insulin resistance and treating diabetes.Methods:The diabetic rats were prepared.The protein activities of InsR phosphorylation and IRS-1 tyrosine phosphorylation were detected by applying Western blotting method.Results:The standard of a mold for the diabetic rats is fasting plasma glucose ≥ 11.1mmol / L and model rats were randomly recruited.The result of fasting blood glucose(FBG) showed that the Jangtang Bushen Prescription can reduce FBG of diabetic rats,the FBG of high-dose of Jiangtang Bushen Prescription group and aspirin group showed a no difference(P0.05).The result of Western blotting method showed that protein activity of InsR phosphorylation in muscle tissue of diabetic rats comparison between the model group and control group showed significant difference(P0.01),the protein activity of InsR phosphorylation in muscle tissue of diabetic rats comparison between low-dose of Jiangtang Bushen Prescription group and aspirin group showed a no difference(P0.05),and the 2 groups were on the same level.The protein activity of IRS-1 tyrosine phosphorylation in muscle tissue of diabetic rats comparison between the model group and control group showed significant difference(P0.01),the protein activity of IRS-1 tyrosine phosphorylation in muscle tissue of diabetic rats comparison with high-dose of Jiangtang Bushen Prescription group,aspirin group and control group showed no difference(P0.05),the high-dose of Jiangtang Bushen Prescription group and aspirin group were on the same level(P0.05).Conclusion:Jiangtang Bushen Prescription can increase the protein expressions of InsR phosphorylation and IRS-1 tyrosine phosphorylation to a certain degree,which may be one of the mechanisms of improving insulin resistance and treating diabetes by inhibition of inflammatory signal transduction pathway.
