摘 要:目的:研究黄芩苷对流感病毒FM1感染小鼠肺组织NO及氧化损伤的保护作用。方法:将96只ICR鼠随机分为正常对照组、模型组、利巴韦林组(100mg.kg-1.d-1),黄芩苷大剂量组(1 500mg.kg-1.d-1),黄芩苷中剂量组(750mg.kg-1.d-1)和黄芩苷小剂量组(375mg.kg-1.d-1),每组16只。用流感病毒亚甲型鼠肺适应株A/FM/1/47(H1N1)感染小鼠,建立小鼠流感病毒性肺炎的生物模型,用不同剂量黄芩苷溶液灌胃治疗后,观察小鼠肺指数、肺组织H-E染色切片病理形态变化,比色法检测肺组织匀浆总NO合成酶(NOS)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-PX)含量或活性。结果:黄芩苷中、小剂量组均能明显降低肺指数,减轻肺组织病变;显著下调肺匀浆总NO合成酶、丙二醛及上调谷胱甘肽过氧化物酶含量或活性(P<0.05,P<0.01)。结论:黄芩苷可通过抑制流感病毒性肺炎小鼠肺组织NO及过氧化物水平而发挥保护作用。Objective: To investigate the protective function of baicalin on lung NO and oxidized damages to influenza virus infection in mice.Methods: 96ICR mice were randomly divided into six groups: normal control,model control,ribavirin(100mg·kg-1·d-2),baicalin high dose(1 500mg·kg-1·d-2),baicalin middle dose(750mg·kg-1·d-2) and baicalin low dose(375mg· kg-1·d-2),16 rats in each group.The pneumonia model was established by infection of influenza virus strain A/FM/1/47(H1N1).Model mice were treated with different dose group of baicalin by intragastric administration.The contents of NOS,MDA and GSH-PX was detected by color methods.At the same time,observing the pathological change of lung injury.Results: middle and low dose groups of baicalin decreased the contents of NOS and MDA,increased the contents of GSH-PX obviously(P0.05,P0.01),reduced lung index,alleviated the pathological change of lung injury significantly.Conclusion: Baicalin has an obviously protective effect by decreasing the level of NO and peroxides in the lungs of mice infected with influenza virus FM1.
