动物种属差异及黏膜部位对灯盏乙素经离体鼻黏膜渗透性的影响

摘　　要：目的:考察动物种属差异及黏膜部位对药物经鼻黏膜渗透性的影响。方法:灯盏乙素为模型药物,采用立式扩散池,分别以新鲜的离体犬、小型猪、家猪离体鼻黏膜为渗透屏障,药物置于鼻黏膜上方,以pH6.8 PBS为接收液,进行渗透试验(n=4),HPLC法测定灯盏乙素。结果:灯盏乙素经犬、小型猪、家猪鼻黏膜渗透系数分别为(16.162±1.957)×10-3cm/min,(13.697±0.605)×10-3cm/min,(4.740±0.211)×10-3cm/min;灯盏乙素经上述动物离体鼻黏膜的渗透系数之间均存在显著性差异。灯盏乙素经离体家猪鼻中膈前部与鼻中膈黏膜的渗透系数分别为(0.194±0.004)×10-3cm/min、(4.740±0.211)×10-3cm/min,两者比较,具有显著性差异。结论:灯盏乙素可经上述离体鼻黏膜渗透,但渗透系数存在种属差异;灯盏乙素经同一动物鼻黏膜不同部位的渗透性亦存在差异。Objective： To study the effects of species differences and the site of mucous membranes on drug＇ permeability through nasal mucosa in vitro.Methods： The Franz diffusion cell method was used to experiment the permeability test（n=4）,taking fresh nasal mucosa of dog,swine and domestica in vitro as permeation barrier separately,with Scutellarin,as the model drug,above them,and blank pH6.8 PBS as absorption liquid.HPLC was used to determine the scutellarin.Results： the permeability coefficient of Scutellarin through nasal mucosa of dog,swine and domestica in vitro are（16.262±1.957）×10-3cm/min,（13.697±0.605）×10-3cm/min,（4.740±0.211）×10-3cm/min,separately.There were significant differences among them.The permeability coefficient of Scutellarin through nasal mucosa of Anterior Spetum and spetum are（0.194±0.004）×10-3cm/min,（4.740±0.211）×10-3cm/min,separately.There were significant differences.Conclusion： Scutellarin could permeate through the above-mentioned nasal mucosa in vitro.There might be different permeability coefficient among different species.The permeability of Scutellarin might also be difference at the different site of mucous membranes.
