摘  要:目的 制备高纯度的糖基化终产物(AGEs) 用作免疫原或检测标准品。方法 牛血清白蛋白(BSA) 与葡萄糖在体外共同孵育制备AGEs - BSA,经凝胶层析纯化后,应用荧光光谱和SDS- 聚丙烯酰胺凝胶电泳鉴定。结果 纯化后AGEs - BSA 与标准的AGEs - BSA 电泳图谱基本一致;激发和发射光谱均为单峰,分别位于360 nm 和446 nm 。结论 应用凝胶层析纯化AGEs - BSAObjective To prepare high purity advanced glycation end products serving as immunogen or standard product.Methods AGEs-BSA was obtained by incubation of BSA with glucose, then purified by gel filtration chromatography and identified by SDS-PAGE and fluorescence spectra.Results The AGEs-BSA purified by us has almost the same picture as standard AGEs-BSA which was purified by perfusion chromatography and the monopeak of the excitation and emission spectra. The peak of the excitation and emission spectra was located at 360?nm and 446?nm respectively.Conclusion The purity of AGEs-BSA purified by gel filtration chromatography was satisfactory.
