摘 要:目的观察水芹提取物(OJE)对小鼠肾阳虚的对抗作用。方法将小鼠随机分成6组,除正常组外,其余各组每日腹腔注射25 mg/kg氢化可的松,正常组每日腹腔注射等量生理盐水,同时,各药物组分别按0.2 mL/10 g给予相应药物灌胃治疗,正常组和模型组用生理盐水灌胃,1次/d,连续给药10 d。测定血清中环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)、性激素、丙二醛(MDA)、超氧化物歧化酶(SOD)水平。结果与正常组比较,模型组雄鼠阳虚症状明显,自主活动次数明显下降(P<0.01),cAMP降低、cGMP升高,cAMP/cGMP比值降低(P<0.05),睾酮浓度明显降低(P<0.05),MDA、SOD明显提高(P<0.05),阳虚模型建立成功;而模型组雌鼠各症状不明显,各指标无明显变化,未形成阳虚模型。与模型组比较,OJE可明显缓解雄鼠阳虚症状,显著提高自主活动次数,提高cAMP,显著降低cGMP,显著提高cAMP/cGMP比值,显著提高睾酮水平,降低MDA浓度,提高SOD活力;而OJE对雌鼠各指标有一定作用,但无统计学意义。结论 OJE可明显治疗由氢化可的松引起的小鼠肾阳虚症状,其作用机制可能与调节cAMP/cGMP平衡、改善机体内自由基状况以及调节性激素水平有关。Objective To explore the role of extract of Oenanthe Javanica (OJE) against the deficiency of kidney yang in mice. Methods The male and female mice were randomly divided into six groups: normal group, model group, positive control group and OJE large, medium and small dose group. The normal group were daily injected intraperitoneally with normal saline, and other groups with 25 mg/kg hydrocortisone. At the same time, normal group and model group were given saline, other groups were given drugs at the dose of 0.2 mL/10 g, once a day, for ten days. cAMP, cGMP, sex hormone, MDA and SOD in the serum were determined. Results Compared with the normal group, male of the model group showed strong symptoms of deficiency of kidney yang, locomotor activity decreased significantly (P〈0.01), cAMP decreased, cGMP increased, cAMP/cGMP ratio decreased significantly (P 〈 0.05), testosterone level decreased significantly (P 〈 0.05), MDA significantly increased (P〈0.05), and SOD significantly improved (P〈0.05). Yang defciency model was established successfully. However, female of the model group showed no obvious symptoms and no significant changes in each index. Compared with the model group, OJE could significantly alleviate the yang deficiency symptoms of male mice, increase locomotor activity, increase cAMP, decrease cGMP, increase cAMP/cGMP, increase testosterone level, lower MDA concentration and increase SOD activity. OJE played a little role in all the indicators of the female mice, but without statistical significance. Conclusion OJE can significantly treat the yang deficiency symptoms of mice caused by hydrocortisone. Regulating cAMP/cGMP balance, improving the status of the body free radicals and regulating the level of sex hormones may be its mechanism.
