摘  要:目的研究经大鼠溃疡皮肤反复给予朱红膏对肾组织丙二醛(MDA)、Na+-K+-ATP酶及金属硫蛋白(MT)的影响,初步探讨朱红膏中汞经皮吸收作用于肾的机理。方法将80只SD大鼠随机分为8组:朱红膏A组(1218.56 mg/kg)、B组(609.28 mg/kg)、C组(304.64 mg/kg)、D组(152.32 mg/kg)、E组(76.16 mg/kg)5个剂量组及基质组(凡士林)、溃疡模型组、皮肤破损组。溃疡模型采用皮肤缺损+埋置异物+细菌感染法造模,皮肤破损组仅以刀片刮伤大鼠皮肤,基质组及朱红膏各剂量组将药纱条直接贴敷于创面,每日给药4 h,连续给药14 d。测定肾组织中MDA含量、Na+-K+-ATP酶活力,并采用酶联免疫分析法测定MT含量。结果与溃疡模型组比较,朱红膏A组MDA含量升高(P<0.05);朱红膏A组Na+-K+-ATP酶活力显著降低(P<0.01),基质组与朱红膏C、D、E组酶活力均呈不同程度的升高;朱红膏A、B、C、D组MT含量均显著升高(P<0.01),其余差异无统计学意义(P>0.05)。结论反复过量使用朱红膏,汞经皮吸收作用于肾脏,可以引起脂质过氧化,影响能量代谢,诱导MT的产生。Ab Objective To study the effects of Zhuhong Ointment on MDA, Na+-K+-ATPase and metailothionein (MT) level of kidney in skin ulcer rats. Methods Eighty SD rats were randomly divided into Zhuhong Ointment A (1 218.56 mg/kg), B (609.28 mg/kg), C (304.64 mg/kg), D (152.32 mg/kg), E (76.16 mg/kg) group, Vaseline group, ulcer model group and impairment control group. Zhuhong Ointment was applied four hours per day on ulcers, which were established on the absence skin, foreign body and staphylococcal infection. The levels of MDA, Na+-K+-ATPase and MT in kidney were tested after administration for 14 days. Results Compared with ulcer model group, MDA content in A group increased significantly (P〈0.05). The level of Na+-K+-ATPase dropped down obviously in A group (P〈0.01), while it increased in Vaseline group and the group C, D and E. The level of MT increased in group A, B, C and D (P〈0.01). Conclusion The renal effects of using excess Zhuhong Ointment repeatedly may lead to oxidant injury and production of Na+-K+-ATPase and MT.
