摘 要:目的探讨肝再生磷酸酶-3小干扰RNA(PRL-3 si RNA)对异位子宫内膜间质细胞骨架的影响。方法选择2009年11月至2010年5月在浙江大学医学院附属妇产科医院行卵巢内异囊肿剔除术的子宫内膜异位症患者32例,原代培养异位子宫内膜间质细胞并鉴定,运用siRNA技术干扰沉默PRL-3基因,Westernblot检测PRL-3蛋白表达,激光扫描共聚焦显微镜观察PRL-3 si RNA对异位子宫内膜间质细胞骨架的影响。结果经PRL-3 si RNA干扰后PRL-3蛋白表达明显下降,与对照组相比较,实验组异位子宫内膜间质细胞中应力纤维数目明显增多、变粗而清晰,而细胞的丝状、板状伪足却明显减少。结论 PRL-3 si RNA能有效沉默PRL-3,并且使异位子宫内膜间质细胞骨架构型显著改变,从而导致异位子宫内膜细胞迁移能力下降。Objective To evaluate the effects of PRL-3 siRNA on the endometriotic stromal cells cytoskeleton by laser scanning confocal microscopy. Methods Endometriotic stromal cells were primarily cuhured and identified in vitro. SiRNA was designed to silence PRL-3 gene, and PRL-3 protein expression was detected by western blot. Then endometriotic stromal cells cytoskeleton was observed by laser scanning confocal microscopy. Results The expression of PRL-3 protein significantly decreased in experimental group, compared with control group. Furthermore, stress fibers became thick and clear in the experimental group, while filaments and pseudopodia plate significantly reduced Compared with control group. Conclusion PRL-3 siRNA could silence PRL-3 gene, make significant changes of endometriotic stromal cells cytoskeleton distribution, which decreased cell migration ability. It suggested that PRL-3 may be a potentially attractive therapeutic strategy in endometriosis.
