摘 要:目的: 建立一种高效液相色谱快速测定人血清中头孢曲松浓度的分析方法。方法: 使用Bio- Rad 高效液相色谱仪, LunaTM C18 (2) 柱(4-6 m m ×150 m m , 5 μm) , 流动相为甲醇- 0-1 % 三乙胺水溶液(15∶85 , 用磷酸调pH 至7-5) , 检测波长为240 nm , 内标为茶碱。结果: 头孢曲松人血清浓度在0-75 ~384 μg·m L- 1 范围内线性良好, 最低检出浓度为0-5 μg·m L- 1 , 日内及日间精密度均优于5 % 。结论: 本方法经济、快速、灵敏、准确、专属性高。Objective:To establish a simple HPLC method for the determination of ceftriaxone in human serum Method: A Bio-Rad 1350t HPLC instrument with Luna TM C 18 (2) (4 6 mm×150 mm,5 μm) column was used The mobile phase was methanol-0 1% triethyl amine aqueous solution (15∶85, pH was adjusted to 7 5 by phosphoric acid).UV detector was set at 240 nm and theophylline was used as internal standard Results: A linear relationship was obtained between the peak height ratio and the concentration of ceftriaxone from 0 75 to 384 μg·mL -1 and the limit of detection of ceftriaxone in serum was 0 5 μg·mL -1 Both intraday and interday precisions were better than 5% Conclusion: The method is economic, rapid, sensitive and accurate
