摘 要:目的: 研究分离鉴别石蒜科生物碱加兰他敏和力可拉敏( 二氢加兰他敏) 及加兰他敏含量测定的方法。方法: 采用硅胶薄层色谱分离鉴别石蒜生物碱, 以二氯甲烷- 甲醇- 氨水(90∶10∶0-5)展开, 稀碘化铋钾试剂显色; 高效液相色谱法采用硅胶柱, 流动相为二氯甲烷- 甲醇(100∶8) , 检测波长为254 nm , 线性范围为80 ~320 μg , r = 0-999 6 。结果: 硅胶薄层色谱可分离鉴别石蒜生物碱加兰他敏和力可拉敏, 硅胶柱高效液相色谱法分离加兰他敏和力可拉敏及测定加兰他敏的含量, 分离效果好, 定量准确。结论: 本法简便快速, 定量准确。在原料药和制剂的分析中有推广价值。Objective:To study the methods for identification and determination of galanthamine, lycoramine and other Amaryllidaceae alkaloids in galanthamine and its preparations Methods:Thin layer chromatography and HPLC method are used for the detection, identification and determination of these alkaloids TLC on silica gel developing by CH 2Cl 2-MeOH-NH 4OH (90∶10∶0 5) and detected with Dragendorff′s reagent and separated by HPLC on silica gel column with mobile phase containing CH 2Cl 2-MeOH (100∶8) at 1 0 mL·min -1 flow rate and quantified with an UV detector operated at 254 nm Results:The tested Amaryllidaceae alkaloids were successfully separated by TLC on silica gel and by HPLC The lycoramine and other alkaloids have no effects on the assay of galanthamine by HPLC Conclusion:The method is of simplicity, sensitivity and accuracy and can be used both for the analysis of galanthamine and galanthamine in drug formulations
