摘 要:目的 通过分析2 型糖尿病患者的前激素转化酶(PC)2 基因多态性探讨2 型糖尿病的发病机制。方法 运用聚合酶链反应和聚丙烯酰胺凝胶电泳方法,分析162 例2 型糖尿病患者和124 名正常人PC2 基因第2 号内含子中串联重复序列(CA) 的多态性与2 型糖尿病发病及病情的关系,并与国外文献报道比较不同人群之间等位基因频率分布的区别。结果 PC2 基因共存在7 种多态性,分别由15 ~21 个(CA)组成,在2 型糖尿病患者中,(CA)21 等位基因频率明显高于正常人群。中国人和日本人2 型糖尿病患者在该等位基因分布频率方面不存在差异,该基因多态性与2 型糖尿病患者的病情无关。结论 在上海地区中国人2 型糖尿病患者中PC2 基因(CA)21 等位基因的频率高于正常人。Objective To investigate the pathogenesis of type 2 diabetes mellitus by analyzing the polymorphism of prohormone convertase (PC) 2 gene of type 2 diabetic patients. Methods 162 type 2 diabetic patients and 124 normal control subjects were selected, the polymorphism of simple tandem repeat (CA) in the second intron of PC 2 gene was analyzed with polymerase chain reaction and polyacrylamide gel electrophoresis. The distribution of the allele frequencies was compared to the results reported, and the relationship between the allele types and the blood glucose control of the patients was also analyzed. Results 7 alleles were found, including 15~21 (CA) tandem repeat. In type 2 diabetic patients, (CA) 21 allele frequency was significantly higher than that in normal control. No difference was found in allele frequency distribution in type 2 diabetic patients between Chinese and Japanese. No relation was found between the polymorphism of PC2 gene and blood glucose control. Conclusions The frequency of (CA) 21 allele of PC2 gene is higher in type 2 diabetic patients than that in normal control in Shanghai Chinese.
