摘  要:目的 探讨大鼠2 型糖尿病时高血糖对磺脲类药物受体(SUR)的影响。方法 采用逆转录聚合酶链反应(RTPCR)技术,在大鼠注射链脲菌素后0、4、12 、24 周取其血和胰腺β细胞分别测定血糖、血脂、SURmRNA 转录水平。结果 在前4 周大鼠的血糖已开始升高,但SUR 的转录水平和对照组相比并无差别;随着血糖的升高和高血糖时间的延长,胰岛β细胞的SUR 表达水平呈下降趋势。结论 在链脲菌素所致2 型糖尿病大鼠模型中磺脲类药物受体在基因转录水平上并无改变。SUR表达水平下降可能为继发性磺脲类药物失效原因之一。Objective To probe into the effect of hyperglycemia on transcription of sulfonylurea receptor (SUR) mRNA in pancreatic tissue of type 2 diabetic rats induced by streptozotocin. Methods The blood glucose,lipid were measured and gene transcription of SUR in β cells of pancreatic tissue of the diabetic rats at 0, 4, 12 and 24 weeks were quantified by using reverse transcription polymerase chain reaction (RT PCR). Results In the first 4 weeks the level of blood glucose began to increase in diabetic rats, but the levels of SUR mRNA transcription were not different between diabetic rats and control rats. With the elevation of blood glucose and prolongation of hyperglycemia, the levels of transcription of SUR were low in β cells of pancreatic tissue of diabetic rats. Conclusion There is no difference in transcription of SUR gene between control group and type 2 diabetes mellitus group. Decreased transcription of SUR mRNA may be one reason of secondary effetiveless of sulfonylureas.
