摘 要:通过对紫外吸收光谱、试剂用量、显色温度等因素进行考查,确定苯酚-硫酸法测定红参单寡糖含量的最佳试验条件。结果表明,5%苯酚1mL、硫酸7mL、60℃保温30min、检测波长490nm时,葡萄糖检测浓度在20~120μg/mL范围内与吸光度呈良好的线性关系(R2=0.999 3),平均回收率100.3%,RSD=2.99%。该方法操作简便,精密度、重现性、稳定性良好,测得加糖红参中单寡糖含量是未加糖红参(对照)的2~4倍,可用于红参外源糖的检测。In order to determine the optimum testing conditions,the wave length,reagents and reaction temperature etc were investigated.Results showed that when the 1.0mL of 5% phenol aqueous solution and 7.0mL of concentrated sulfuric acid were used to complete the reaction at 60℃,glucose had a good liner relationship(R2=0.993) with absorbency ranged from20 to 120μg/mL,and with an average recovery rate of 100.3%,RSD= 2.99%.Total content of oligosaccharides in red ginseng (added sugar) was 2~4 times higher than CK.This method,with good precision,reproducibility and stability,was simple and convenient,and was suitable for detection of red ginseng foreign sugar.
