摘 要:目的探讨电针影响骨性关节炎软骨细胞JAK-STAT信号通路表达的作用机制。方法采用成年SD大鼠60只,抽签法随机分为正常组、造模组,造模组采用4%木瓜蛋白酶双膝关节腔注射建立骨性关节炎模型,抽签法随机分为模型组、对照组、实验1组、实验2组、实验3组,实验组采用电针干预,检测关节液中TGF-β1含量,Real time RT-PCR检测软骨细胞STAT3、Smad3、LepR mRNA的表达。结果电针治疗后关节液中TGF-β1含量明显高于模型组(P<0.01),关节软骨中STAT3、Smad3、LepR mRNA表达均明显高于模型组(P<0.01,P<0.05)。结论电针有效增加TGF-β1含量及促进关节软骨中STAT3、Smad3、LepR mRNA的表达,可能是其防治骨性关节炎的重要机制之一。Objective To observe the effect of electroacupuncture on JAK-STAT signal transduction pathway in chondrocyte of osteoarthritis rats and explore its mechanism.Methods 60 adult SD rats were divided randomly into normal group,model group,control group,experimental group 1,experimental group 2 and experimental group 3,the osteoarthritis models were established by intra articular injection of 4% papain and rats in experimental group were treated with electroacupuncture.The content of TGF-β1 insynovial fluid was detected,and the mRNA expressions of STAT3,Smad3 and LepR in cartilage were detected by Real time RT-PCR.Results The content of TGF-β1 in synovial fluid of rats in each experimental group was significantly higher than that of model group after electroacupuncture(P0.01),and the mRNA expressions of STAT3,Smad3 and LepR in cartilage of rats in each experimental group were significantly higher than those of model group(P0.01,P0.05).Conclusion Electroacupuncture can increase the content of TGF-β1 in synovial fluid effectively and promote the mRNA expressions of STAT3,Smad3 and LepR in cartilage,which may be one of the mechanisms of treating osteoarthritis.
