摘  要:目的建立透骨消痛方汤中芍药苷、迷迭香酸的含量测定方法。方法采用高效液相色谱法,色谱柱:Phenomenex Gemini C18(250×4.6 mm,5μm);流动相:乙腈为流动相A,0.2%的磷酸为流动相B,梯度洗脱;流速:0.8 mL/min;检测波长:芍药苷为233 nm,迷迭香酸为330 nm;柱温:35℃。结果芍药苷回归方程为Y=12.591X+53.691(r=0.9991),在12~240μg/mL范围内线性关系良好,平均加样回收率为99.59%,RSD为0.16%;迷迭香酸回归方程为Y=30.713X-212.62(r=0.9993),在6.012~60.12μg/mL范围内线性关系良好,平均加样回收率为99.01%,RSD为0.45%。结论 2种成分的峰都能有效分离,其测定方法准确、重复性良好,方中其它成分对测定无干扰。Objective To establish a HPLC method for determination of paeoniflorin and rosmarinic acid in Tougu Xiaotong recipe.Methods The separation was performed on Phenomenex Gemini C18(250×4.6 mm,5 μm) with gradient elution of acetonitrile-0.2% phosphoric acid solution as mobile phase,flow rate was 0.8 mL·min-1,the detective wavelength of paeoniflorin,rosmarinic acid was 233 nm,330 nm respectively,and the column temperature was 35℃.Results The linear ranges of paeoniflorin and rosmarinic acid were 12~240 μg·mL-1(r=0.9991),6.012~60.12 μg·mL-1(r=0.9993) respectively,the average recoveries were 99.59%(RSD=0.16%),99.01%(RSD=0.45%) respectively.Conclusion The peaks of the two ingredients could be segregated effectively.The method is accurate and reproducible,and there is no interference to the result from other ingredients in the recipe.
