摘  要:为了明确玉米弯孢叶斑病菌的侵入机理,对农杆菌介导法(ATMT)转化弯孢叶斑病菌的条件进行了探究。以农杆菌AGL-1为侵染菌系、pCAMDGFP作为转化载体。结果表明,玉米弯孢叶斑病菌的ATMT最佳转化条件为:分生孢子萌发12 h后,与农杆菌在25℃黑暗条件下共培养48 h,共培养介质为微孔滤膜。转化效率可达到100~110个转化子/106个孢子,并获得转报道基因GFP的弯孢叶斑病菌单孢。In order to analyze the Curvularia lunata ingression mechanism,this study explored the main factors affecting Agrobacterium-mediated Curvularia lunata.The recombinant vector pCAMDGFP was transformed in this study into fresh conidia of Curvularia lunata via Agrobacterium tumefaciens(AGL-1).The results showed that germination time of Curvularia lunata conidia was 12 h at 25 ℃;co-culturing time of the germinated conidia and bacteria was 48 h at 25 ℃;co-culturing media was millipore filter;and the transformation frequency was up to 100-110 transformants per 106 conidia.Transgenic GFP Curvularia lunata spores were also generated in this study.
