摘 要:目的优化推理选育条件,获得头孢菌素C高产菌株。方法根据头孢菌素C的生物合成途径和代谢调控机制设计菌种推理选育方案,原始出发菌株经紫外诱变后,采用托盘琼脂柱筛选法依次对铜离子、丙二酸及头孢菌素C耐受菌株进行选育。结果最终获得一株头孢菌素C高产菌株TBC-68071,其产量较原始菌株提高了154.6%,且高产特性稳定。结论对铜离子、丙二酸及头孢菌素C耐受的顶头孢霉菌株可以高产头孢菌素C,推理选育克服了随机筛选的盲目性,提高了菌种筛选的效率。Objective To Optimize conditions of rational selection and obtain cephalosporin C high-producing strains. Methods An efficient rational selection method were designed according to the biosynthetic pathway of cephalosporin C and mechanism of metabolic regulation. After UV mutagenesis, cephalosporin C high-producing strains were screened by an agar column high-throughput method with the pressures of copper ions, malonic acid and cephalosporin C tolerance. Results The yield of cephalosporin C of the final selected strain TBC-68071 was increased 154.6% than the original strain, and the high-producinggenetic characteristics was stable. Conclusion The results showed that rational screening ofAcremonium chrysogenum overcame the blindness of random screening and improved the screening more efficiently.
