化脓灸预处理对肝纤维化大鼠肝损伤保护作用的实验研究

摘　　要：目的:通过制备免疫损伤性肝纤维化大鼠模型,探讨化脓灸预处理肝纤维化大鼠肝损伤的保护作用,为进一步研究化脓灸抗肝纤维化作用提供实验依据。方法:于造模时对动物进行化脓灸预处理,检测肝组织TIMP-1和MMP-2基因表达量定量分析、肝组织匀浆羟脯氨酸(Hyp),观察肝组织病理变化。结果:化脓灸能显著降低肝纤维化大鼠的TIMP-1mRNA水平(P<0.05),但对MMP-2mRNA指标作用不明显。同时能明显降低肝纤维化大鼠肝组织匀浆羟脯胺酸(Hyp)水平,显著减轻肝脏细胞的病理损伤。结论:化脓灸具有有一定的保肝护肝作用,有助干预防和阻断肝纤维化的发生,其机制可能与抑制TIMP-1mRNA的基因表达水平有关。Objective： Through preparing liver fibrosis model rats with immune injury,to probe the protecting function of pretreatment of festering moxibustion on liver injury of rats with liver fibrosis,to provide experimental basis for further study on the anti-hepatic fibrosis of festering moxibustion.Methods： The model rats were given a pretreatment of festering moxibustion,the quantitative analysis of gene expression of liver tissue TIMP-1 and MMP-2 and the liver tissue hydroxyproline（Hyp） were detected,and the hepatic pathological changes were observed.Results： The festering moxibustion can significantly reduce TIMP-1 mRNA level of liver fibrosis（P0.05）,but it has no obvious effects on MMP-2mRNA.At the same time it can significantly reduce the hydroxy proline（Hyp） level of liver fibrosis rat,and significantly reduce the pathological damage of liver cells.Conclusion： The festering moxibustion has a certain protecting function on liver,and helps to prevent and block the occurrence of liver fibrosis,and its mechanism may be related to the inhibition of the level of gene expression of TIMP-1mRNA.
