摘 要:目的:探讨粉防己碱(Tet)对血管性痴呆(VaD)大鼠学习记忆能力和海马神经元凋亡的影响。方法:将40只大鼠随机分为假手术组、模型组、Tet干预组(含10和30mg/kg两个剂量组),采用双侧颈总动脉永久性结扎(2-VO法)建立VaD大鼠模型。造模12周后进行Morris水迷宫实验检测大鼠行为学,HE染色观察海马CAl区神经元病理学变化,利用TUNEL原位末端标记法检测海马神经元凋亡情况。结果:与模型组相比,Tet干预组大鼠Morris水迷宫实验逃避潜伏期明显缩短(P<0.05),穿越平台次数明显增多(P<0.05);HE染色发现Tet干预组,海马CAl区锥体细胞病理改变较模型组有明显改善:Tet干预组海马CA1区神经元凋亡数目较模型组明显减少(P<0.05)。结论:粉防己碱(Tet)通过抑制海马CA1区神经元凋亡,能明显提高VaD大鼠的学习能力和空间记忆能力,发挥对神经元的保护作用。Objective:To observe the effect of tetrandrine on learning and memory and the neuronal apoptosis in the CA1 zone of hippocampus in rats with vascular dementia(VaD).Methods: Model group rats were performed by permanent bilateral common carotid artery ligation(2-VO).Morris water maze test was adopted to test ability of learning and memory in all rats 12 weeks after surgery.HE staining and TdT mediated dUTP nick end labeling(TUNEL) were used to detect neuronal apoptosis.Results: Escape latency was significantly shorten and the number of entries in the target area was significantly increased in rats administrated with Tet when compared with those in model group(P0.05).At the same time,numbers of the TUNEL positive cells was significantly decreased in the CA1 zone of hippocampus in Tet-treated rats(P0.05).Conclusion: Tet could improve the learning and memory,and reduce the quantity of the TUNEL positive cells in the CA1 zone of hippocampus of rats with vascular dementia.
