HIF-1α在骨髓间充质干细胞治疗大鼠急性心肌梗死中的作用

摘　　要：目的观察低氧诱导因子-1α(HIF-1α)在骨髓间充质干细胞(BMMSCs)治疗大鼠急性心肌梗死中的作用,并与单纯BMMSCs移植作比较。方法①BMMSCs的提取、培养、标记。②建立急性心肌梗死模型。将大鼠随机分为4组,对照组、单纯BMMSCs组、HIF-1α反义寡核苷酸组、HIF-1α错义寡核苷酸组。术后4周,测定大鼠左室收缩压、舒张末压、压力变化最大上升与最大下降速率,检测细胞移植情况及新生血管密度。结果①大鼠血流动力学检测:与对照组、HIF-1α反义寡核苷酸组相比,BMMSCs组与HIF-1α错义寡核苷酸组左室收缩压、压力变化最大上升与最大下降速率均明显升高,舒张末压明显降低。②心肌超微结构检测:BMMSCs组与HIF-1α错义寡核苷酸组大鼠心肌超微结构损伤少于其他两组。③心肌血管密度:BMMSCs组与HIF-1α错义寡核苷酸组大鼠梗死心肌内血管密度高于其他组。结论 HIF-1α反义寡核苷酸对BMMSCs治疗急性心肌梗死具有抑制作用,BMMSCs对大鼠急性心肌梗死的部分治疗作用可能是通过HIF-1α介导的。Objective To observe the effect of hypoxia-inducible factor-1α （HIF-1α） on bone marrow mesenchyreal stem cells （BMMSCs） treatment in rats with myocardial infarction （MI）. Methods The rats were randomly divided into four groups after operation： MI control group, BMMSCs group, antisense oligonueleotide for HIF-1α group and scramble oligonucleotide for HIF-1α group. Four weeks after operation, LVSP, LVEDP, ＋ dp/dtmax,-dp/dtmax were detected, and capillary densities were measured in ischemic musculature. Results （1） Compared with control group, BMMSCs group and scramble oligonucleotide for HIF-1α group, LVSP, ＋ dp/dt - dp/dtmax were lower, while LVEDP was higher in antisense oligonucleotide for HIF-1α group. （2） The myocardial ultrastructure： the dgree of myocardial uhrastructure injury was minor in BMMSCs group and scramble oligonucleotide for HIF-1α group compared with the other groups. （3） Survived BMMSCs and capillary densities were decreased in the antisense oligonucleotide for HIF-1α group. Conclusion The results suggest that antisense oligonucleotide for HIF-1α inhibited the effects of BMMSCs in the therapy of MI in rats. The therapeutic effect of BMMSCs may be regulatede by HIF-1α.
