摘 要:目的探讨维甲酸(RA)在体外诱导大鼠胚胎垂体生长激素(GH)细胞的分化作用。方法原代培养大鼠胚胎垂体细胞,采用不同浓度的维甲酸(10-8、10-7、10-6、10-5 mol/L)诱导大鼠胚胎垂体细胞分化,以DMEM培养基为作为对照组,诱导2、4、6 d后,利用免疫组化检测GH细胞百分比,放射免疫分析检测细胞上清GH分泌量。MTT法检测维甲酸(10-6 mol/L)或碱性成纤维生长因子(bFGF,10-6 mol/L)对大鼠胚胎垂体细胞的增殖活性。结果免疫组化和放射免疫分析结果显示:诱导6 d后,10-6 mol/L维甲酸能够提高GH细胞的百分比和GH分泌量(均P<0.05)。MTT检测发现:bFGF可显著促进胚胎垂体细胞增殖(P<0.05),维甲酸组细胞增殖活性未见明显增加(P>0.05)。结论维甲酸能够在体外诱导胎鼠垂体GH细胞分化,具有一定的量效和时效关系。Objective To explore effect ofretinoic acid (RA) in inducing the differentiation of fetal rat pituitary somatotroph cells in vitro. Methods Primary fetal rat pituitary cells were cultured and treated with different concentrations of RA (10-8, 10-7, 10-6, 10-5 mol/L) to induce the differentiation of growth hormone (GH) cells, and the cells treated with DMEM served as the control group. Immuno- histochemistry was used to detect the percentage of GH ceUs and radioimmunoassay to detect secretion of GH in supernatant 2, 4, 6 d after the treatment. The proliferation of fetal rat pituitary cells was evaluated by MTT assay after treatment with RA (10-6 mol/L) and basic fibroblast growth factor (bFGF, 10-6 mol/L). Results Immunohistochemistry and radioimmunoassay results showed that RA ( 10-6 mol/L) increased the percentage of GH cells and secretion of GH (both P〈0.05) 6 d after the treatment. MTT assay indicated that 10-6 mol/L bFGF promoted the proliferation of fetal rat pituitary cells (P〈0.05), while there was no obvious increase in 10-6 mol/L RA group (P〉0,05). Conclusions RA can induce the differentiation of fetal rat pituitary somatotroph cell in vitro with relations of dose-effect and time,effect.
