摘  要:目的研究CarO蛋白与鲍曼不动杆菌耐环丙沙星的相关性。方法收集长沙地区四家医院的鲍曼不动杆菌56株进行药敏实验、对临床敏感株进行环丙沙星体外诱导耐药实验、PCR扩增CarO基因并测序、实时荧光定量PCR比较临床敏感株和耐药株的CarO基因表达情况。结果所收集的临床鲍曼不动杆菌对环丙沙星的耐药率达85.7%,临床敏感株被诱导成耐环丙沙星菌株后,其CarO基因序列发生了改变,且CarO基因表达量明显降低。结论 CarO蛋白表达下降与鲍曼不动杆菌耐环丙沙星有一定的相关性。【Objective】 To study the relationship between carbapenem-associated resistance Omp(CarO) and quinolones-resistant Acinetobacter baumannii.【Methods】 Clinical Acinetobacter baumannii isolates were collected from four hospitals in Changsha to study antimicrobial susceptibility testing.Some clinical sensitive strains were induced into ciprofloxacin-resistant strains by the successive selection in medium containing increasing concentrations of ciprofloxacin.The CarO gene was proliferated by polymerase chain reaction(PCR) and the PCR products were sequenced.The gene expression of CarO mRNA was analyzed by real-time PCR.【Results】 Of all collected Acinetobacter baumannii isolates,85.7% were ciprofloxacin-resistant strains.After the sensitive strains were induced into resistant ciprofloxacin,their CarO gene got some difference,and the gene expression of CarO mRNA declined.【Conclusions】 The decline of CarO protein expression in Acinetobacter baumannii is associated with ciprofloxacin resistance to some extent.
