补肾活血方对血管性痴呆小鼠脑组织BDNF和bFGF含量的影响  被引量:5

摘  要:目的探讨补肾活血方、补肾方、活血方对血管性痴呆(VD)小鼠学习记忆能力及脑组织脑源性神经营养因子(BDNF)、碱性成纤维细胞生长因子(bFGF)含量的影响。方法采用反复脑缺血再灌注法复制VD小鼠模型,以尼莫地平为阳性对照组,观察补肾活血方、补肾方、活血方三组小鼠水迷宫法行为学及脑组织BDNF、bFGF含量的变化。结果与假手术组比较,模型组小鼠学习与记忆能力下降,表现为游全程时间延长,错误次数增加(P<0.01),脑组织BDNF和bFGF含量明显升高(P<0.05或P<0.01);与模型组比较,各治疗组学习成绩和记忆成绩均有明显提高(P<0.05或P<0.01),脑组织BDNF和bFGF含量明显升高(P<0.05或P<0.01);各治疗组比较,补肾活血组的学习成绩、记忆成绩及脑组织BD-NF含量优于补肾组、活血组和阳性对照组(P<0.05或P<0.01),脑组织bFGF含量高于活血组和阳性对照组(P<0.05)。结论补肾活血方可通过提高脑组织BDNF和bFGF含量,改善VD小鼠学习与记忆能力,且补肾活血方在改善学习记忆能力和提高BDNF含量方面优于活血方和补肾方,提高bFGF含量优于活血方。Objective To explore the effect of bushenfang,huoxuefang,and bushenhuoxuefang on ability of learning and memory,and the content of basic fibroblast growth factor(bFGF) and brain-derived neurotrophic factor(BDNF)in brain tissue of vascular dementia(VD) mouse.Methods Operation of cerebral ischemia reperfusion was used to reproduct the VD model,and the praxiological changes were observed by the means of water maze and the contents of bFGF and BDNF in brain tissue were observed.Results Compared with normal group,the ability of VD model mouse for learning and memory were reduced,which showed extending the time for swimming the whole process,and increasing times of mistakes(P〈0.01),and the contents of bFGF and BDNF were increased(P〈0.05 or P〈0.01).Compared with model group,the learning and memorizing performance record were improved(P〈0.05 or P〈0.01),and the contents of bFGF and BDNF were increased(P〈0.05 or P〈0.01) in each treatment group;the learning and memorizing performance record and the contents of BDNF of the bushenhuoxue group were better than those of other treatment group(P〈0.05 or P〈0.01),the content of bFGF of the bushenhuoxue group was better than that of huoxuefang(P〈0.05).Conclusions The bushen,huoxue,and bushenhuoxue traditional Chinese medicine could increase the contents of bFGF and BDNF,and improve the learning and memory capability.Bushenhuoxue is better than bushenfang and huoxuefang in improving learning and memory and increasing BDNF,and better than huoxuefang in increasing bFGF.
