摘 要:建立新的中低压制备色谱技术,分离纯化茜草蒽醌单体。先采用高压液相色谱技术分析所分离产物的纯度,再用核兹共振谱、红外、质谱分析确证单体的化学结构。结果表明:用中低压色谱分离茜草的乙醇提取物而得到的4个单体,其纯度分别为99.87%、99.56%、99.42%、98.21%,结构鉴定为从石油醚萃取物中分离得到的单体为大叶茜草素,从氯仿萃取物中分离得到的单体为1,3,6-三羟基-2-甲基蒽醌,从乙酸乙酯萃取物中分离得到的单体分别为1,3,6-三羟基-2-甲基蒽醌-3-O-(3′,6′-O-二乙酰基)-α-鼠李糖(1→2)-β-D-葡萄糖苷和1,3,6-三羟基-2-甲基蒽醌-3-O-新橙皮糖苷。A new technique of middle and low pressure preparative chromatogram was established, and the anthraquinone monomer from rhizoma of Rubia cordifolia were separated and purified. Purity of the compounds was analyzed by HPLC,and the structures of the compounds were confirmed by MS. 1 ^1H-NMR and 13C-NMR. IR. The result showed that monocases were successfully obtained from ethanolic extract of Rubia cordifolia, with purities of 99. 87% ;99.56% ; 99. 42 % ;. 98. 21%. Mollugin was separated from petroleum ether extract; 1,3, 6-tribydroxy-2- methyl anthraquinone was separated from chloroform extract; 1,3,6- trihydroxy- 2-methyl anthraquinone- 3-0- (3'6'- C-acetyl)-a-rhamnose(1→2)-β-D- glucoside and 1,3,6-trihydroxy-2-methyl anthraquinone-3-0- neohesperidoside were separated from acetic ether extract.
