摘  要:研究正在生长烟株单个叶片黑暗诱导衰老过程,并对叶片的形态结构、化学成分及衰老相关基因表达和酶活变化进行分析。结果表明,被处理叶片逐渐由绿变黄直至褐色,衰老相关基因CP1、CLH表达逐渐增强,CAB、RBCS2B基因表达逐渐减弱,这些基因表达水平与作为对照的正常衰老的叶片相应时期基本一致。超微结构观察发现,黑暗处理叶片中的叶绿体中嗜锇颗粒增多增大,类囊体片层模糊并降解;叶片物质代谢,酶活变化与正常衰老叶片中变化规律相似。因此,黑暗处理烟叶能有效快速诱导衰老,其衰老模式与正常烟叶衰老模式基本一致。We studied the regularity of tobacco senescence,which was induced by individually dark treatment.Also,the changes of leaf morphology,mesophyll cell ultrastructure,chemical ingredients,senescence-related genes expression and enzymatic activities were analyzed in the individually darkened leaves.The results showed that the color of tobacco leaf which was induced by individually dark treatment turned yellow and brown,and the ultra structure of the mesophyll cell in the leaves indicated that the osmiophilic globule became more and bigger,the lamellae turned blurred and then degraded.While the level of senescence-related genes expression,enzymatic activities and chemical ingredients in different treatment time were similar with the control check leaves(CK) which grown at the normal condition.Consequently,tobacco senescence could be induced by dark treatment rapidly and efficiently,and the mode of which was analogous to the normal senescence.
