摘 要:以美国黑核桃、核桃楸、青林核桃实生子代及部分普通品种共86份种质资源为试验材料,利用8对SSR引物进行遗传多样性分析及聚类分析。结果表明,8对黑核桃引物共扩增出48条带,其中多态性带44条,占总扩增带的91.67%;遗传多样性指数为0.197 2~0.473 4,Shannon指数为0.304 3~0.666 3。说明黑核桃微卫星在核桃近缘种中能得到有效扩增,可以用于近缘种遗传多样性分析和核桃属种间关系研究。聚类结果表明,青林实生子代具有丰富的遗传多样性,与核桃楸及黑核桃具有明显差异,但与其他普通品种界限不明显。Eight simple sequence repeat(SSR) primer pairs of black walnut were used to study the genetic diversity of Juglans nigra of American,J.stenocarpa Maxim.and Qinglin walnut(Juglans regia L.) populations and other cultivars(total of 86 samples).The results show that totally 48 DNA fragments were amplified with 8 simple sequence repeat(SSR).The high polymorphic DNA fragments were 44,accounting for 91.67 % of all.The distribution of genetic diversity index was 0.197 2~0.473 4 and the distribution of Simpson index was 0.304 3~0.666 3.The black walnut microsatellites can be effectively amplified in closely related species in the walnut and can be used for analyzing the genetic diversity in related species and relationship in other Juglans species.The cluster results indicated that Qinglin walnut has relatively larger genetic diversity with Juglans mandshurica and Juglans nigra,but closely related to the other cultivated species.
