单鼻腔通气障碍小鼠模型嗅黏膜的组织学变化

摘  要:目的建立单鼻腔通气障碍的小鼠动物模型,并观察其嗅黏膜的组织学变化情况。方法取刚出生的昆明小鼠只,显微镜下缝合小鼠左侧前鼻孔。在刚出生,出生后2周、4周、6周时分别取5只小鼠头部制成石蜡冠状位切片。以嗅标记蛋白(OMP)特异抗体对切片行免疫组织化学染色,对比观察小鼠两侧鼻腔嗅黏膜的厚度和嗅标记蛋白表达强度。结果刚出生时小鼠两侧鼻腔嗅黏膜厚度、嗅标记蛋白表达强度大致相同(P>0.05)。第2周时,左侧鼻腔嗅黏膜略薄、嗅标记蛋白表达强度略低,但差异不显著(P>0.05)。第4周时,这种差异变得显著(P<0.05)。第6周时,这种差异变得非常显著(P<0.01)。结论显微镜下缝合小鼠前鼻孔,可以作为构建单鼻腔通气障碍小鼠动物模型的可靠方法。鼻腔通气障碍单一因素即可以使嗅黏膜萎缩,成熟嗅感觉神经元的数量明显下降。[ Objective ] To establish mouse model with dysfunction of unilateral nasal ventilation and observe histological changes of its olfactory epithelium, [Methods] Twenty newborn Kunming mice were selected to sew up their left anterior naris under microscope. Five of them were made into coronal paraffin sections of their heads every two weeks. These sections were detected by Immunohistostaining with OMP (Olfactory Marker Protein) antibody. The thickness of olfactory epithelium and intensity of OMP expression of bilateral nasal cavities were observed and compared. [ Results ] The thickness of olfactory epithelium and intensity of OMP expression were almost same when mice were just born. After two weeks, the thickness of olfactory epithelium of left nasal cavity was a little thinner and the intensity of OMP expression was a little less than that of right nasal cavity. The differences were not obvious (P 〉0.05). After four weeks, the differences were obvious (P 〈0.05). After six weeks, the differences were very obvious (P 〈0.01). [ Conclusions] It is a reliable method to establish mouse model with dysfunction of unilateral nasal ventilation by sewing up anterior naris under microscope. Dysfunction of nasal ventilation may cause atrophy of olfactory epithelium and obvious decrease of mature ORNs independently.
